Nimesulide is an NSAID and a preferential COX II inhibitor. It is a potent analgesic, antipyretic, and potent Anti-Inflammatory action.
Pharmacokinetic:
It behaves as a competetive inhibitor of histamine release in-vivo and in-vitro, hence posseses antihistaminic and antiallergic properties.
Nimesulide does not have significant gastroduodenal side effects. The onset of analgesic and antipyretic action of oral nimesulide is 1.6–3.17 h and duration of action is 10–12 h.
Absorption:
Well absorbed from GI tract following oral admin. Peak plasma levels:1-3 hours. With bid admin of 100 mg, steady-state is achieved within 24-36 hours.

- Nimesulide has less gastrointestinal adverse effects than other NSAIDs
- It has superior benefit-risk ratio to other drugs with a comparable safety and tolerability to placebo.
- Its multifactorial mode of action gives it a unique and broad action on inflammatory.
- Nimesulide is also safe in sensitive asthmatic patients. It is reported to be beneficial in relieving the symptoms of rhinitis, rhinopharyngitis tubaritis and seceretory otitis media with concomitant antibiotic treatment.
